Research in the Department of Biochemistry
The Department has made some contribution to the scientific and pedagogical personnel, only the last 10 years at the department graduate students, 15 doctoral applicants are protected and about 60 Ph.D. theses. The main scientific directions of the department over the years were: clinical biochemistry, environmental biochemistry and toxicology, patohimiya thermal injuries, the biochemical mechanisms of action of medicinal compounds. Clinical chemistry before studying the molecular mechanisms of pathogenesis and pathogenetic treatment of diseases of various organs and systems: the cardiovascular system, gastrointestinal tract, nervous system, musculoskeletal system, hemorrhagic fever with renal syndrome and other viral diseases, neoplastic process. A series of papers was devoted to the study patohimii trauma and thermal injuries, tuberculosis, Biochemistry kumys, microorganisms, hypovitaminosis, intrauterine infection in newborns and other
In this case, for example, unknown earlier links in the pathogenesis of hemorrhagic fever with renal syndrome (HFRS). Shown that the major role in the pathogenetic mechanisms of development, the progression of hemorrhagic and renal syndrome, other metabolic and functional abnormalities in this disease have intensified lipid peroxidation, characteristic changes in the exchange of prostaglandins, components of the hemostatic system, nitric oxide production, changes in cytokine status and hemorheologic properties of uniform elements of blood.
Revealed that the unidirectional shift in the form of increasing lipid peroxidation, depression of antioxidant protection, activation of intravascular coagulation and fibrinolytic system failure, metabolic disorders, products of polyunsaturated fatty acids – the suppression of depressor and pressor hyperproduction series prostanoids are integral components of the pathogenesis of HFRS, and pronounced dysfunction of these systems is observed the development of its complications. It is shown that the formation of DIC-syndrome in this disease an important role changing ability of red blood cells to aggregate in violation of erythrocyte membranes, decreased activity of membrane ATPase and the accumulation of erythrocyte lipid peroxidation products. Based on these data are developed and introduced new technologies of treatment and rehabilitation of patients with HFRS.
In summarizing the research on thermal injuries can note the following. Department staff, the regularity of changes of metabolism of proteins, lipids and carbohydrates, energy metabolism and hormonal status in burn disease in childhood. It is experimentally shown that thermal injury leads to shortage of power supply of fabrics with a reduction in content and production of ATP in cells, a violation of the processes of tissue respiration, synthesis of pyridine and the ratio of oxidized and reduced forms, characterized by the appearance of metabolic blocks dichotomous glucose oxidation and decrease in its tissue utilization, and activation processes of gluconeogenesis, an increase in energy value of fatty acids, glycerol, amino acids. Found that a significant prevalence of catabolic reactions on anabolic protein metabolism, leading to the development of protein deficiency, mainly due to increased permeability of lysosomal membranes with the activation of tissue proteases and induction of tumors. It was found that for severe burns is characterized by changes in hormonal status with inhibition of production of thyroid hormones, androgens and increasing levels of ACTH, cortisol, endogenous opioid peptides, glyukogona and insulin, a violation of hormone-receptor interactions, inhibition of insulin binding, decreased cytosolic androgen receptors. Research in clinical state of metabolism in children with thermal injuries proved the applicability of the developed in the experiment approaches, which justify and implement a kambustologiyu new techniques and treatments that have proven in the treatment of children after the “burn” the disaster train station at Ulu Telyakovsky in 1989
Studies on the biochemical mechanisms of action of medicinal compounds in our department were laid by the works of one of the Head of Department Prof. IL Vakulenko. In this direction were also performed dissertations, MA Abdulmeneva, FH Kamilov, EA Molodtsova, AS Yuhanovoy, RM Kondratenko, AR Halimova, ES Volkova, IE Abramova. Pharmacological correction of metabolic shifts in various states of special chapters of theses: GH Mirsaevoy, VB Yegorova, GA Pavlova, AN Zakirova, RM Salyahovoy, RA Salakhova, EM Nizamova, AD Akhmetov I.Zh. Shaybakova, AV Perevalova, FH Shaymuhametovoy, NT Gabdullin, etc. It is possible to identify a series of works carried out in conjunction with pharmacists under the guidance of Head of Department of Pharmacology № 1 Honorary Academician, Academy of Sciences of Belarus, Laureate of State Prize, Professor DN Lazarus, according to the metabolic mechanisms of pharmacological effects of pyrimidine derivatives – pentoxyl, methoxy (methyluracil), oksimetiluratsila etc.
The leading scientific direction of the department especially in recent years, along with clinical biochemistry, is the environmental and toxicological chemistry. In this direction the Department is actively cooperating with several academic institutions of Bashkortostan and Russia.
Studied the toxicological and metabolic effects of new organic products and compositions that are embedded in the aerospace and aviation industries, as well as aromatic and organochlorine compounds, the largest producer in Russia which is of Bashkortostan. Experimentally studied the laws of development of metabolic disorders under the influence of herbicides based on 2,4-dichlorophenoxyacetic acid, chlorinated biphenyls, piromeelitovogo anhydride and other compounds belonging to the group of persistent organic pollutants.
As a result, studies show that in the development of intoxication under the influence of these pollutants are of prime importance increased microsomal oxidation with an induction of synthesis of RNA, cytochrome p450 proinformatsionnoy and b5, and increasing concentrations of monooxygenase enzymes of brain tissue, liver, lungs, kidneys, and the activation of free radical oxidation and peroxidation of membrane lipids, impaired transport and receptor proteins of cell membranes, reduced physiological antioxidant protection. The level and severity of these changes depends on the frequency and intensity of the proceeds of persistent organic pollutants. At the same time in the cells shows a shift of processes of tissue respiration and energy production. The dynamics of changes in ATP, energy charge of cells, the processes of energy production and utilization of ATP is characterized by a certain pattern: at low (subthreshold and threshold) concentrations of toxicants or their non continuous immersion flow cell power system compensates for the existing workload, although there are changes in the ratio of aerobic and anaerobic processes, the intensity Synthesis and perefosforilirovaniya adenine nucleotides shift the concentrations of major electrolytes in the extra-and intracellular spaces. Under the action of higher concentrations or prolonged admission toxicants notes development ATP deficit with an uncompensated, although reinforced its synthesis. Simultaneously, a decrease in the level of enzymatic and enzymatic antioxidant protection, synthetic phase of detoxification, the growth of electrolyte imbalance, impaired metabolism of nucleic acids, protein, glycogen, lipids and other phenomena of metabolic decompensation. The method of Northern-dot molecular hybridization of the organs of rats with labeled DNA fragments of genes confirmed that the introduction of chlorinated biphenyls and dioxins as well as the stimulation of gene transcription is precisely toksifitsiruyuschey isoforms of cytochrome P-450-CYP 1A1, which was associated with the processes of mutagenesis, teratogenesis, carcinogenesis, changes in the metabolism of steroid compounds, and others assumed that the inducing effect of these halogenated pollutants on CYP 1A1 mediated cytoplasmic ariluglevodorodnym receptor.
Polarographic method, using radioactive substrates demonstrated in liver mitochondria of experimental animals with the introduction of organochlorine pollutants reduction of protein biosynthesis, efficiency of oxidative phosphorylation during the oxidation of NAD-dependent substrates and relatively intact area flavinzavisimogo respiratory chain. Revealed a reduction in activity of some enzymes citrate cycle and glutamate.
Targeted studies of the effect of chlorinated biphenyls (sovtol Sovol) and fenoksigerbitsidov (amine salt of 2,4-dichlorophenoxyacetic acid) on organs and systems have provided new data reveal their effect on the blood system, immune system, as well as their hypothyrosis and demaskuliniziruyuschego effects. Thus, reveal the mechanism of development of hematological disorders accompanying intoxication by herbicides. It is shown that the animals are observed a dose-dependent on the severity and time of development of hemolytic changes, characterized by accelerated destruction of red blood cells as a result of destabilization of their membranes, inhibition of erythropoiesis in bone marrow and, as a consequence, the development of true anemia, with a reduction in functional and metabolic activity of leukocytes, extensive morphological and functional disorders of various organs and tissues. Revealed that the effect immunotropic fenoksigerbitsidov characterized suppressive effect on the humoral, decreased energy and plastic processes in the macrophages with the subsequent change in their functions: antigen, clearance, Fe-assay, phagocytosis. It is shown that under conditions of toxic chlorinated POPs developed marked changes in thyroid status, which reflects the active elimination iodtironinov of blood flow, slowing down their metabolism and reduce peripheral hormonal thyroid function and is characterized by insufficient blood concentrations of thyroid hormones, the predominance of metabolically inactive counterparts. Simultaneously, the signs of morphofunctional thyroid gland, the molecular mechanisms tireoidzavisimyh disorders of energy metabolism of hepatocytes, the activity of intracellular enzymes tireoidzavisimyh. One of the main reasons for developing intoxication hlorfenoksigerbitsidami hypothyrosis as demaskuliniruyuschego effects is desensitization of the competent cells of the hypothalamic-pituitary system to the central and peripheral regulatory influences. It is shown that the basis of changes in gametogenesis and androgenogeneza, biochemical and morphological parameters of semen and sperm during intoxication POPs are the inhibition of oxygen consumption and the main substrate oxidation, changing the ratio of oxidized and reduced pyridine coenzymes, inhibition of enzymes of energy and plastic metabolism, enzyme ensembles biosynthesis of sex steroids in the testes, the prevalence of free radical oxidation processes with a decrease in antioxidant protection, reducing the sensitivity of cells of the reproductive organs to stimulate testosterone. Gonadotropic effects fenoksisoedineny are also characterized by suppression spermatopoietic testicular activity involving in pathological process mainly mature gametes. Reduce fertilizing properties of semen during intoxication hlorfenoksigerbitsidami attributable to lower sperm concentration, restrictions on their mobility, respiratory activity and viability, inhibition of fructose consumption, change in the activity of marker enzymes of gametes, reducing the protein content and glucose, the lack of testosterone and prolactin.
The studies have provided, firstly, to formulate a common molecular mechanism of the negative actions of this group of chemical factors of maladjustment, to clarify the sequence of interrelated biochemical reactions that leads to the formation of the pathological process after prolonged entry of small doses of toxicant – Development premorbitnogo state.
Secondly, the results revealed a complex and justify informative biochemical indicators of early diagnosis prenosological manifestations of the pathogenic effects of the studied POPs (chlorinated biphenyls, fenoksigerbitsidy, piromelitovy dianhydride and some cyclic products of its process of synthesis), which is of interest in a monitor health assessment of persons with Work and non contact with this group of compounds, as well as indicators to assess the negative impact of complex environmental toxicants on the health of workers refining, petrochemical and chemical industries and the urban population with severe environmental stress and trouble.
Thirdly, have been identified and experimentally proved methods and schemes of pharmacological correction of developing metabolic and functional changes, and recommendations for prevention prenosological changes in the body under the action of anthropogenic pollutants. So, given the significant reduction of retinol and tocopherol in the blood of animals with chronic poisoning hlorfenoksigerbitsidom was the correction of biochemical changes with use of vitamins A and E. presupposed scheme was effective – contributed to the normalization membrane stability, the level of TBA-reactive products in erythrocytes and methemoglobin kinetics ferritsianidnogo education. In acute and chronic intoxication chlorinated biphenyls shown antioxidant, membrane stabilizing and hepatoprotective effects oksimetiluratsila possessing immunomodulating and anabolic effect. The action of chlorinated biphenyls bromantan installed protective effect associated with modulation of synthesis of pre-mRNA of cytochrome P450 1A1 in the URA cells of liver and lung gene URA 1A2 in the liver and cytochrome b5 gene in cells of various tissues. Shown that the introduction of exogenous thyroid hormones during intoxication with chlorinated herbicides and chlorinated biphenyls promotes the normalization of the liver decreased activity of enzymes of energy metabolism – succinate -, malate -, glutamate, limiting enzymes mochevinoobrazovaniya – karbamoilfosfatsintetazy and arginase, an increase in oxygen consumption in all metabolic states of mitochondria and the positive shift in the energy performance of hepatocytes.
In order to confirm the experimental data were carried out targeted research on indicators of biochemical, immunological and hormonal status, health workers major occupational groups of chemical plants for the production of herbicides based on 2,4-dichlorophenoxyacetic acid, pyromellitic dianhydride and some cyclic compounds – products of its synthesis, urban with an unfavorable ecological situation.
Studying the health of children living in conditions of combined effects of toxicants, has shown its dependence on the intensity of technogenic pollution of residence. In areas with higher levels of environmental stress is defined higher overall incidence of pre-school children, found lower levels in the blood of carotene, retinol and α-tocopherol, reduction of the peroxide resistance of erythrocytes, a significant drop in the number of catalase, increasing concentrations of insulin, estradiol, a strong variability of the content cortisol, lower triiodothyronine and thyroxine.
Determination of hormones in the blood of more than 2000 men of reproductive age in urban areas with high concentration of petrochemical and chemical industries (Ufa, Sterlitamak, Salavat) revealed a significant shift of their content, compared with the reference values. Thus, the average content of the main sex hormone – testosterone found in 1,5 – 2 times lower concentration of gonadotropic hormones differed considerable fluctuations, but on average did not differ at the same time observed hyperprolactinemia, giperestradiolemiya, dissociation levels of ACTH and cortisol. The findings suggest that demaskuliniziruyuschey and feminizing orientation adjustment processes of the endocrine regulation of reproduction for men in conditions of ecological stress. Generalization of experimental studies, clinical examination of men, the prevalence of the male reproductive system diseases, laboratory data and biochemical analysis of semen, metabolic and functional state of spermatozoa to propose the concept of reproductive health of men as a sensitive indicator of environmental well-being of the region. Based on these results, the principles of monitoring male fertility under the influence of environmental factors, the amendments to the Law on Reproductive Health of the Republic of Bashkortostan. Studies of production workers fenoksigerbitsidov OAO Ufahimprom also revealed changes in sexual content, gonadotrophic hormones and thyroid hormones, lipid metabolism disturbances with increased cholesterol and atherogenic lipoproteins, the development of metabolic changes characteristic of the acceleration of biological aging: the immune deficiency with a reduction in T-lymphocytes , levels of immunoglobulins, depression function of phagocytic cells associated with metabolic disorders: decrease in cellular content of glycogen and lipids, an increase of catabolic processes and lipid peroxidation, the decline in antioxidant protection.
Workers productions pyromellitic dianhydride in the blood plasma, erythrocytes and urine of the indices, reflecting an intensification of lipid peroxidation in erythrocytes increased activity of antioxidant enzymes: catalase, peroxidase, superoxide dismutase, glutathione peroxidase, glutathione, reduced the content of α-tocopherol, koferola varies nature of chemiluminescence of urine and blood plasma. In erythrocytes, depending on the duration and intensity of the production of contact with the studied aromatic compounds decreased energy charge, the level of Ca, Mg – dependent ATPase. Based on these data have been developed recommendations for the prevention of the negative impact of production factors on the health of workers by improving working conditions, intensity and duration of work experience, employment of workers these industries.
Naturally, this is not a complete list of scientific studies conducted at the department, but just the highlights and major achievements. Significantly more problems, probably waiting for us ahead.
History of the Department of Biochemistry
One of the first in the Institute was in 1932, organized by the Department of Organic Chemistry. The first teacher of organic chemistry was adopted MA Abdulmenov. In 1933, organized by another 2 chair of chemical-sky profile: Biological Chemistry and fizkolloidnoy chemistry.
The first lecturer of biological chemistry was enlisted to September 1, 1933 Ibrahim Zileevich Ilyasov , who worked previously as an assistant Oren-burg Institute of beef cattle.
Department of Organic and Biological Chemistry in 1933 are located on the second floor of this building No 2 University Str. Frunze, 47. As the head of the Department of Biological Chemistry, was invited to the penny-chair of Biological Chemistry, Saratov State University-theta Eugene M. Gubarev . December 31, 1933 People’s Commissar of Health of the RSFSR, including Professor EM Gubarev was approved by the head of the Department of Biochemistry BGMI. He was awarded the title of professor. February 7, 1934 Professor EM Gubarev started work as head of the Department fizkolloidnoy and biological chemistry. In 1934, as assistant of the department was adopted Rudolph Emilievich Wever.
In 1935, assistant of the department of biochemistry was adopted by Ivan Logginovich Vakulenko , a former assistant professor and then professor of Biological Chemistry, Tomsk State University.
In the same year the decision of the Qualification Commission of the People’s Commissariat-care he was awarded the degree of Doctor of Medical Sciences, he was confirmed as the title of professor, and from 7 February 1936, he moved to the management department of organic chemistry.
In 1937, the Department of the buildings on the street. Frunze 47, moved into a rented building, called the Chemical Corps. It was a two-story stone building with central heating, which was on Verhnetorgovoy square, 41. This building housed the department of biological, organic and fizkolloidnoy chemistry and inorganic chemistry with a course of analytical chemistry.
In 1937, the department had taken 2 more assistant: SM Bar-Jonah on biological and MG Rakhmankulov on fizkolloidnoy chemistry. Later, the Department added to assistant MZ Batalov. In the same year assistant IZ Ilyasov was transferred to a common chemistry, Acting Head of Department.
Formation of the Department of Organic Chemistry, Bioorganic and fizkolloidnoy went long and rather complicated way. Each of the first teachers made a great contribution to the organization, formation and development departments.
Research focused on the biochemistry department of vitamins and micro-organisms. Organization of the experimental laboratory allowed Professor Gubareva in 1939 to complete and defend a thesis for the degree of Doctor of Biological Sciences on the topic “Effects of formaldehyde on proteins.
In 1940, assistant SM Ionin defended his thesis for the degree of Candidate of Biological Sciences on “The Influence of salt factor in terms of thermal coagulation of milk.” Assistant IZ Ilyasov prepared and passed in 1941 to protect a Dissertation Council of the Tomsk State Medical Institute, the thesis for the degree of candidate of biological sciences on the theme “Some indicators of metabolism in clinical scurvy.” Young employees of chemical departments are actively conducted their first scientific research.
Over the years, has been improved material base of the chairs. The Department received a new, built and put into operation in 1940. Biofiziologicheskom housing spacious rooms.
In 1941 he completed the spring exams, and on June 22 the war started. All the men chairs, including professors, have entered into a people’s militia received military training, and other staff members studied and passed the standards GSO and PVHO. Out the front assistants MZ Batalov, MG Rakhmankulov, IZ Ilyas. MZ Battles in the same year died, in 1942, M. F. Rakhmankulov missing.
Institute in July 1941 freed biofiziologichesky Corps, and in October – student hostel number 2 to hospitals. The department was once again time-meschena in the building on ul. Frunze, 47. In late October, on the basis of the institute was evacuated to Ufa posted 1-st Moscow Medical Institute with faculty and students of a total of 600 people. Two of the Institute within 2 years of training exercises conducted in the same premises in two shifts. The planned training sessions at the department violated separation of students to help industrial enterprises, agriculture, military hospitals and the very institution.
Despite the difficulties associated with the wartime conditions, the Department staff continued to work successfully and honorably fulfilling educational, methodical, scientific and educational work.
At the time, needed a major restructuring of the educational and methodical work. Steel is widely practiced methods of students’ independent work. Since April 1942g.kafedra Organic Chemistry joined the department of biological chemistry tion. The head of the joint Department of Biological and Organic Chemistry of the order of the Institute was appointed Professor EM Gubarev, who led it until 1945, Professor IL Vakulenko, he headed the department of organic chemistry in 1935 -1941 GG, became the second professor.
A notable is the fact that the head of the course of physical and colloid chemistry in 1941-1943 GG the department served as Vice – President of the Academy of Sciences of the Ukrainian SSR and the director of the Institute of Biochemistry, Academy of Sciences USSR, Academician AV Palladin. As one of the pioneers vitaminology in the USSR, he became in these years, the creator of water-soluble analog of vitamin K – vicasol. Under his leadership, developed the technology for manufacturing and industrial Company now HYDRATED issue at the Ufa Vitamin Plant methylnaphthochinon, and somewhat later and vicasol (vitaminaK3). The drug was widely used in in-ennyh hospitals in the rear and the front. This has been facilitated and carried out in Ufa, 15-17 June 1943 conference, whose participants expressed their appreciation to both drugs, especially vicasol as factor replacement stopping various parenchymal and capillary bleeding, which stimulates wound healing. In 1942, AV Palladin was elected a member of the USSR, in 1944 – Academy of Medical Sciences, in 1946-1962 GG worked as the President of the Academy of Sciences of USSR, in 1955. awarded the title Hero of Socialist Labor. In 1964-1972 GG was president of the All-Union Biochemical Society.
The war made changes in the plan of scientific research department. Professor EM Gubarev completed prior scientific research defense of a thesis for the degree of Doctor of Medicine on “Chemical and immunological properties of carbohydrates and proteins on the Scientific Council 1-st Moscow Medical Institute in 1942 (UFA). Along with the systematic research on the biochemistry of microorganism the body naturally, works dedicated to the immunological and chemical properties of proteins and carbohydrates diphtheria bacilli, EM Gubarev with Prof. NL Vakulenko and employees Clinic of Nervous Diseases (Head, Professor N. Savchenko) performed a study on metabolic changes in patients with closed brain injury, in traumatic neuro-Zach wartime. Proceedings of the departments’ Materials in a closed brain injury in the deep stages of the evacuation “was of great practical importance for determining the degree of disability at the shell-shocked. This scientific work was awarded the III degree of the editorial board of The Experience of Soviet Medicine in Great Patriotic War of 1941-1945”.
In 1944, Bashkortostan suffered a great tragedy. As a result, consumption of food grains wintered outbreak of septic sore throat, from which wiped out entire villages. As a result of assistance provided by the Government of the republic of the USSR, managed to overcome the disease. However etiopato-genesis, prevention and treatment of septic angina (the modern name of the disease) have been studied enough. In the same year in the journal Biochemistry, published an article EM Gubareva “Chemical nature of the poison that causes septic sore throat”, which summed up the hard work of staff, Department of Biochemistry. The results of this work in 1945 were published in a thematic Proceedings of the Institute of septic tonsillitis.
For various and fruitful work during the war-tion of honorary titles “Honored Worker of Science BASSR” was awarded both a professor of IL Vakulenko, and EM Gubarev.
Department after the release biofiziologicheskogo corps returned to their rooms on the first floor of the building. This spacious room with a usable area of 375 square meters was before March 1980
In connection with the transition of Professor EM Gubareva to charge the Department of Biochemistry, Rostov Medical Institute in 1945, was elected the chair of Prof. IL Vakulenko.
After the war, returned to college, many teachers. October 9, 1945, arrived at the Department of demobilized assistant Ilya Petrovich Kelyuh, who participated not only in the Great Patriotic War, but also in the Civil War in the battles against the armies of Wrangel. In 1946, after demobilization, he returned assistant IZ Ilyas. – Major Corps of Engineers, in the same year he successfully defended his dissertation for the degree of candidate of biological sciences, placed in Tomsk Medical Institute in 1941.
Since 1946 the department was carried out teaching of the three disciplines: biology, organic chemistry and fizkolloidnoy. Lectures on Organic Chemistry professor read IL Vakulenko, on biological chemistry – Associate Professor IZ Ilyasov on fizkolloidnoy Chemistry – Associate Professor Robert E. Wever. State Department consisted of professors, two associate professors and five assistant: IP Kelyuha, MA Abdulmeneva, SG Khamidov, Ph.D., SM Ionin, MP Kuznetsov, senior laboratory MK . Tronin, ZM Tuktarovoy, two lab technicians and senior preparator.
R. E. Wever at a meeting of the Scientific Council defended the thesis for the degree of Candidate of Medical Sciences on the topic: “Effect of salt-free diet on the exchange of certain minerals in patients with tuberculosis of the skin. In 1951 he defended his thesis for Ph.D. in Pharmacy Assistant MA Abdulmenov on “Getting the herbal medicines and dosage forms of European ungulates. In 1948, Robert E. Wever received an academic title of associate professor, and MA Abdulmenov – in 1953
Topic research department was devoted to biochemistry of microorganisms, changes in blood biochemical parameters in a number of diseases – tuberculosis, cancer, etc. As a result of development work done at the department, to detect early forms of cancer have suggested a reaction-la-Kelyuha Vakulenko, based on the allocation of separate fraction of serum glycoproteins.
For achievements in scientific, pedagogical, methodological, educational and scientific-organizational activity in 1947, Professor IL Vakulenko was awarded the honorary title “Honored Scientist of the RSFSR.
At the age of 75 years IL Vakulenko left the management of the department and until his death in 1955 remained the scientific consultant.
